Don’t try to kill them straight away. When you hit a Hoker it shoots spikes that stick to walls and floors. Hit those spikes and they drop a small orb that contains a spine core .

Best way to farm (keeps it simple)
Each Hoker can only take three hits before it dies. So my routine is: hit a Hoker once, wait for it to throw spikes again, then hit the spikes. Repeat.
This gets you multiple cores from the same enemy instead of wasting it by killing it fast. Be quick though, the spikes vanish after a few seconds so if you wait too long you miss the orb. It’s a simple timing thing, nothing fancy.
